AB The lymph node cells and peripheral blood lymphocytes of lymphoma
patients were analyzed for cellular DNR retention, response to efflux
blockers and expression of human differentiation antigens. Extensive
heterogeneity in cellular DNR retention was seen in lymphocytes in 10/66
patients. Cellular DNR retention could be significantly increased by
coincubation with 25 uM prochlorperazine (6/10), 10 uM verapamil (2/10)
or 20 uM dipyridamole (1/10). Analysis for differentiating antigen
expression showed baseline CD2, CD3, CD8 and CD25 counts significantly
associated with increased survival of HIV patients while CD4, CD4 subset
and B cell counts were not. Furthermore, patients with lymphocytes of
CD8 counts greater than 1000 responded to chemotherapy suggesting that
CD8 counts may be an important predictor for HIV associated lymphoma.
These studies underscore the importance of cellular markers in diagnosis
of lymphoma and suggest the use of appropriate efflux blocker or drug
retention and a cytokine for CD8 cell enrichment.
